Background Unmethylated CpG-DNA is a potent ligand for the endosomal Toll-like-receptor-9, important immune activation to pathogen-associated molecules. 1 CpG and other TLR-ligands show effective immunotherapeutic capacities in cancer treatment by inducing an antitumorigenic immunity. 2 They are able reduce tumor progression reduction of intratumoral secretion immunoregulating chemokine CCL22 3 subsequent recruitment immunosuppressive regulatory T cells (Treg), which express CCR4 only so far known receptor CCL22. 4 Our recent work has shown that dendritic (DC) lymph node, mediates tolerance DC-Treg contacts. 5 Indeed, absence CCL22, responses vaccination were stronger resulted rejection. 6 Therefore, we aiming investigate effects on systemic levels, elucidating all involved mechanisms identify new targets immunotherapy. Materials Methods T, B CD11c + DCs wildtype (wt) RAG1 -/- mice isolated from splenocytes magnetic-activated cell sorting vitro assays. Different co-cultures incubated with GM-CSF, as inducer. For vivo experiments, wt treated CpG, R484 or poly(I:C) alone combination GM-CSF. CCL22-levels number organs analyzed. Results Analyzing different compartments , found whole secrete during culture while DC cultured showed no secretion. When reduced splenocytes, it was induced alone. The same results seen when RAG lack functional cells, CpG. suppress unlike Co-cultures however, strongest suppression DC. In could TLR ligands tested significantly. Tumor-associated macrophages promote prostate cancer migration through activation of the CCL22–CCR4 axis
Previous studies have found that tumor-associated macrophages (TAMs) promote cancer progression. We previously reported that TAMs promote prostate cancer metastasis via activation of the CCL2-CCR2 axis. The CCR4 (receptor of CCL17 and CCL22) expression level in breast cancer was reported to be associated with lung metastasis. The aim of this study was to elucidate the role of CCR2 and CCR4 in p...

15 صفحه اولBACH2 represses effector programmes to stabilize Treg-mediated immune homeostasis - a new target in tumor immunotherapy?
Through their functional diversification, distinct lineages of CD4+ T cells play key roles in either driving or constraining immune-mediated pathology and anti-tumor immune responses. Transcription factors are critical in the generation of cellular diversity, and negative regulators antagonistic to alternate fates often act in conjunction with positive regulators to stabilize lineage commitment...
متن کاملEstimation of the Domain of Attraction of Free Tumor Equilibrium Point for Perturbed Tumor Immunotherapy Model
In this paper, we are going to estimate the domain of attraction of tumor-free equilibrium points in a perturbed cancer tumor model describing the tumor-immune system competition dynamics. The proposed method is based on an optimization problem solution for a chosen Lyapunov function that can be casted in terms of Linear Matrix Inequalities constraint and Taylor expansion of nonlinear terms. We...
متن کاملProtein expression profiling identifies cyclophilin A as a molecular target in Fhit-mediated tumor suppression.
Loss of fragile histidine triad (Fhit) expression is often associated with human malignancies, and Fhit functions as a tumor suppressor in controlling cell growth and apoptosis, although specific signal pathways are still undefined. We have used a proteomic approach to define proteins in the Fhit-mediated tumor suppression pathway. Because substitution of Tyr(114) (Y114) with phenylalanine (Y11...
